Verucopeptin (BS-1152) is a cyclodepsipeptide with notable antitumor potency, directly taregts ATP6V1G, a subunit of the vacuolar H+-ATPase (v-ATPase).
Verucopeptin exhibited strong inhibition of both v-ATPase activity and mTORC1 signaling, leading to substantial pharmacological efficacy against SGC7901/VCR cell proliferation.
Verucopeptin inhibited tumor growth SGC7901/VCR xenograft model (1 mg/kg) without significant body weight loss or gross signs of toxicity.
Verucopeptin showed excellent antitumor activity against K562R cells with IC50 of 388 nM, showed substantial inhibition of v-ATPase activity at 10 nM in vitro.
