Biological Activity

WZS0347 is a potent, selective first-in-class PROTAC degrader of transcription factor MITF with potency of 78% degradation at 0.1 uM in B16F10 melanoma cells.
WZS0347 (1 uM) induced concentration-dependent MITF degradation in malignant melanoma cell lines (A375 and GAK).
WZS0347 promoted MITF degradation through a CRBN- and proteasome-dependent pathway.
WZS0347 downregulated MITF and its downstream target genes, suppressing melanoma cell migration.
