Wiskostatin is a specific, cell-permeable chemical inhibitor of the neural Wiskott-Aldrich syndrome protein (N-WASP), inhibits N-WASP-mediated actin polymerization.
Wiskostatin interacts with a cleft in the regulatory GTPase-binding domain (GBD) of WASP in the solution structure of the complex.
Wiskostatin blocks N-WASP activity by stabilizing its autoinhibited conformation, thus leading to a significant decrease in the rate of actin polymerization.
