Biological Activity
XW-17 is a highly potent, selective PARP14 inhibitor with IC50 of 3.03 nM.
XW-17 shows minimal activity against the other tested PARP family members, demonstrating a selectivity index exceeding 9900 over PARP1.
XW-17 induces a concentration-dependent upregulation of PARP14 protein expression, with an EC50 of 59.5 nM in interferon-γ (IFN-γ) stimulated CFPAC-1 cells.
XW-17 inhibits PARP14-mediated ADP-ribosylation in CFPAC-1 and RAW264.7 cell lines.
XW-17 (100 mg/kg) significantly attenuated skin lesions and effectively decreased the expression of key inflammatory factors, including IL-4, IL-13, IgE, and IL-17A, in DNCB-induced AD-like mouse model.
