Biological Activity

XY-280 is a potent, selective, brain penetrant inhibitor of Interleukin-1 receptor-4 (IL-1R4, ST2) with IC50 of 5.85 uM.
XY-280 significantly suppressed the release of PGE2, IL-1β, and IL-6 in LPS-stimulated primary microglia cells.
XY-280 inhibited the ST2/IL-33 signaling in primary microglia cells and suppressed the downstream NF-κB activation.
XY-280 (20 mg/kg, i.p.) markedly reduced mRNA levels of key inflammatory cytokines and chemokines in LPS-induced neuroinflammation model.
