YT-6-2 (YT 6-2) is a small molecule p62/SQSTM1 agonist and ligand, binds to the ZZ domain of the autophagic cargo receptor SQSTM1/p62, exerts antimicrobial efficacy.
YT-6-2 induces the biogenesis and recruitment of autophagic membranes to intracellular bacteria via SQSTM1, leading to lysosomal degradation.
YT-6-2 enhances host innate immunity against Mtb and other pathogens via SQSTM1-mediated xenophagy.
YT-6-2 (5-10 uM) dramatically inhibits the growth rate of Mtb in Mtb-infected BMDMs in CFU assays.
YT-6-2 (i.p. 20 mg/kg) enhance xenophagy-mediated host defense against pathogens in mice.
YT-6-2 inhibits the production of proinflammatory mediators in vitro and in vivo.
YT-6-2 also could be used as p62/SQSTM1 ligand for AUTOTAC design.
