Z-Tyr-Ala-CHN2 is a potent cathepsin L inhibitor with EC50 of <50 nM, has broad-spectrum coronavirus antiviral activity.
Z-Tyr-Ala-CHN2 showed activity against SARS-CoV-1 and HCoV-229E, with EC50 of 0.050 and 0.069 µM, respectively.
Z-Tyr-Ala-CHN2 potently inhibit infection in SARS-CoV-2 both A549-hACE2 and HeLa-hACE2 cells with EC50 of 0.046 and 0.006 uM, with no effect in Caco-2 cells.
Z-Tyr-Ala-CHN2 acts on the endosomal entry pathway in pseudotyped SARS-CoV-2 neutralization assay in VeroE6 cells.
