ZL192 is a small molecule FGF13 ligand / activator and a positive modulator of FGF13/Nav1.7 complex, stabilizes FGF13/Nav1.7 CTD assembly with EC50 of 28 uM, sensitizes Na+ currents in hIPSC-derived sensory neurons.
ZL192 has no agonist activity against the FGF13R110A/Nav1.7 complex.
ZL192 binds to both FGF13 at Arg110 and Nav1.7 CTD at M1830, binds to FGF13 with SPR KD of 4 μM.
ZL192 exerts regulatory effects on Nav1.7 currents only in HEK-Nav1.7 cells coexpressing FGF13 with EC50 of 8.5 uM.
ZL192 and FGF13 overexpression sensitize Na+ currents in hIPSC-derived sensory neurons and are pronociceptive, without triggering stimuli.
ZL192 exhibits pronociceptive roles that are prevented by in vivo FGF13 silencing and recapitulated in FGF13 overexpression.
