ZN148 (APC148) is a specific, irreversible, zinc-chelating metallo-β-lactamase (MBL) inhibitor with broad activity against Ambler class B enzymes, including NDM, VIM, and IMP, restores the in vitro and in vivo activity of meropenem against multidrug-resistant MBL-producing Enterobacterales.
ZN148 (APC148) potentiates the effect of meropenem against NDM-1-producing Klebsiella pneumoniae in a murine neutropenic peritonitis model.
ZN148 shows no inhibition of the human zinc-containing enzyme glyoxylase II at 500 uM.
ZN148 restored the bactericidal activity of meropenem against an NDM-1-producing K. pneumoniae strain.
ZN148 potentiates the activity of meropenem in vivo with no acute toxicity in vivo.
