Biological Activity

Zagociguat (CY6463) is a potent, selective, brain-penetrant and orally-active soluble guanylate cyclase (sGC) activator with EC50 of 66 nM in cell-based cGMP asssays.

Zagociguat (CY6463) stimulates the phosphorylation of CREB in a concentration-dependent fashion in rat primary neurons with EC50 of 25.7 nM.

Zagociguat (CY6463) elicits decreases in blood pressure in both rats and mice.

Zagociguat (CY6463) improves cognitive performance in a model of cognitive disruption induced by the administration of a noncompetitive N-methyl-D-aspartate (NMDA) receptor antagonist.

Zagociguat (CY6463) increases long-term potentiation (LTP) in hippocampal slices from a Huntington's disease mouse model and decreases the loss of dendritic spines in aged and Alzheimer's disease mouse models.
