Biological Activity
dHuR-2 is a molecular glue degrader of human antigen R (HuR), exhibits selective degradation of HuR and CRBN neosubstrates ZFP91 and ZMYM2 with DC50 of 3.8 nM, 1.8 nM and 2.1 nM respectively in Colo205 cells.
dHuR-2 shows CRBN-binding affinity with HTRF IC50 of 0.16 uM,
dHuR-2 shows no effection on GSPT1 and Hu protein paralogues (HuB, HuC and HuD).
dHuR-2 drives CRL4-CRBN-mediated HuR polyubiquitination.
dHuR-2 exhibits selective efficacy in BRAF-mutant CRC and suppresses MAPK signalling.
dHuR-2 modulates VEGFA expression and splicing to suppress tumor angiogenesis.
dHuR-2 inhibits BRAFi-resistant cells and synergize with BRAFi, overcomes EGFR-mediated BRAF inhibitor resistance.
