Biological Activity
eIF2B inhibitor Compound A-(S) is a small molecule stabiliser and allosteric inhibitor of inactive state of eIF2B, shows stabilisation effect of the eIF2B:(P)eIF2-NTD interaction with EC50 of 2.2 uM, stimulating the Integrated Stress Response (ISR).
Compound A-(S) binds the ISRIB pocket of eIF2B, favouring the I-state.
Compound A-(S) binds to eIF2B in complex with phosphorylated eIF2.
Compound A-(S) stabilises the eIF2B decamer and favours eIF2B:eIF2(αP) complex formation.
