pNPS14 is a small molecule inhibitor of phospho-BRD4 (pBRD4) with IC50 of 3.39 uM, binds to phospho-NPS (pNPS) and disrupts HPV E2-BRD4 interaction, effectively inhibits HPV ecDNA replication.
pNPS14 is a small molecule inhibitor of phospho-BRD4 (pBRD4) with IC50 of 3.39 uM, binds to phospho-NPS (pNPS) and disrupts HPV E2-BRD4 interaction, effectively inhibits HPV ecDNA replication. pNPS14 inhibits HPV genome amplification and ori replication but not genome maintenance. pNPS14 binds to phosphorylation-dependent interaction domain (PDID) of BRD4 with Kd of 12.9 uM. pNPS14 shows selectivity toward inhibition of HPV but not host DNA replication. pNPS14 disrupts BRD4 association with 53BP1 and BARD1 impairs differentiation-dependent HPV genome amplification. pNPS14 selectively disrupts 53BP1 and BARD1 binding to HPV ori without reducing corresponding BRD4-L/S occupancy in differentiated W12E cells.
