(S)-3,4-DCPG ((S)-3,4-Dicarboxyphenylglycine) is a potent and selective mGlu8 receptor (mGluR8) agonist with EC50 of 31 nM in cell-based assays, with no acitivity against other mGlu receptors (IC50>3,500 nM).
(S)-3,4-DCPG depressed the fast component of the dorsal root-evoked ventral root potential (fDR-VRP) giving a biphasic concentration-response curve showing EC50 of 1.3 uM.
(S)-3,4-DCPG activates metabotropic glutamate receptors on primary afferent terminals in the neonatal rat spinal cord.
(S)-3,4-DCPG (100 mg/kg) significantly increased c-Fos expression in several stress-related brain regions: paraventricular nucleus of the hypothalamus, central nucleus of the amygdala, lateral parabrachial nucleus and locus coeruleus in mice.
