Biological Activity

2-Methoxyestradiol (2ME2, NSC-659853) is a metabolite of estradiol-17β and a novel antimitotic, antiangiogenic agent, binds to tubulin at or near the colchicine site and inhibits the polymerization of tubulin in vitro.
2-Methoxyestradiol suppresses microtubule dynamic instability in living interphase cells with mitotic IC50 of 1.2 uM, inhibits bFGF and VEGF-induced neovascularization and suppresses the growth of human breast carcinoma in mice.
2-Methoxyestradiol also downregulates hypoxia-inducible factor-1 (HIF-1) at the posttranscriptional level and inhibits HIF-1-induced transcriptional activation of VEGF expression.
