6-OAU (GTPL5846, 6-n-Octylaminouracil) is a potent, specific agonist of GPR84 with EC50 of 512 nM in [35S]GTPγS binding assay, without activity for GPR109A and OXER1 in a panel of 69 GPCRs.
6-OAU activates human GPR84 in the presence of Gqi5 chimera in HEK293 cells with EC50 of 105 nM in PI assays, does not activate human GPR40 in HEK293 cells.
6-OAU provokes chemotaxis of PMNs prepared from human peripheral blood with EC50 of 318 nM, amplifies LPS-stimulated production of the proinflammatory cytokine IL-8 from PMNs and TNFα from macrophages.
6-OAU raises the blood CXCL1 level in rats.
