AAD777 (NVP-AAD777, ZK202664) is a potent, selective VEGFR-2 kinase inhibitor with IC50 of 0.65 uM, weakly inhibits VEGFR-1 and 3 with IC50 of 2.2 and 3 uM.
AAD777 displays little or no activity against PDGFR-β tyrosine kinases, and no activity against FGFR-1, FGFR-3, FGFR-4, PDGFR-α, and Tie-2 tyrosine kinases at 10 uM.
AAD777 inhibits VEGF-induced cellular receptor phosphorylation with IC50 of 26.6 nM in cell-based assays, inhibits VEGF-induced HUVE cell proliferation with IC50 of 19.6 nM.
AAD777 enhances cigarette smoke (CS)-induced oxidative stress, causing augmented inflammatory cell influx and proinflammatory mediators release in mouse lung.
