AAPK-25 is a small molecule inhibitor of DLGAP5 with a binding energy of −9.2 Kcal/mol, suppresses proliferation of PRAD cells via inhibiting DLGAP5/E2F1 axis, also is a potent, dual Aurora/PLK coinhibitor. AAPK-25 effectively inhibited the proliferation of PC3 and DU145 cells with IC50 of 467 nM and 356 nM respectively. 40 nM AAPK-25 significantly hindered the colony formation ability of LNCaP and C4–2 cells, downregulated DLGAP5, E2F1, and ENO2. AAPK-25 (25 mg/kg) suppresses tumor growth and DLGAP5/E2F1 pathway in murine PRAD xenograft model using BALB/c nude mice bearing PC3 cells.
