AB-00011778 is a small molecule inhibitor of SARS-CoV-2 entry and the Spike/ACE2 interaction, inhibits viral replication with EC50 of 250 nM in A549-ACE2 cells, AB-00011778 is equally effective in impairing the S/ACE2 association in the different mutants.
AB-00011778 interferes with the formation of the RBD/ACE2 complex, reaching 50% inhibition at 25 uM in AlphaLISA assays.
AB-00011778 inhibits the binding of S-RBD(His)6 to ACE2 with KD of 111 nM in biolayer interferometry (BLI) assays.
AB-00011778 blocks the viral replication of four major circulating forms of SARS-CoV-2 (the original Wuhan strain and the alpha, delta, and omicron viruses) in pulmonary A549-ACE2 cells with IC50 of 0.5-1 uM, with cytotoxic concentration (CC50) values >30 uM.
