AC1115 is a potent, selective, next-generation antiviral targeting SARS-CoV-2 Mpro and host lysosomal cysteine protease cathepsin L (CTSL) with high antiviral activity, AC1115 is the active form of Olgotrelvir.
AC1115 displays potent inhibition of both wild-type Mpro from the WA-1 strain and Omicron Mpro with IC50 of 2.44 and 14.27 nM, respectively.
AC1115 also displays potent reversible inhibition of human CTSL activity with IC50 of 0.06 nM, >40-fold selective over other members of the human cysteine cathepsin family.
AC1115 showsno significant inhibition against a panel of 56 human and virus proteases.
AC1115 displays antiviral activity against SARS-CoV-2 VOCs, including WA-1, Alpha, Beta, Delta, Lambda, and Gamma variants with EC50 of 0.28-4.26 μM.
