ADX-47273 is a potent and selective mGlu5 positive allosteric modulators (PAM) with with EC50 of 170 nM.
ADX-47273 causes a concentration-dependent increase in the response to 300 nM glutamate in primary astrocyte cultures with EC50 of 230 nM.
ADX-47273 displays no agonist, antagonist, or allosteric responses against rat mGlu1, human mGlu2 and mGlu4.
ADX-47273 increases ERK and CREBP phosphorylation in hippocampus and prefrontal cortex, demonstrates antipsychotic-like and procognitive activities in vivo models.
