Biological Activity
ASM inhibitor 4i (ASM-IN-1) is a potent, selective inhibitor of acid sphingomyelinase (ASM) with IC50 of 1.5 uM.
ASM inhibitor 4i (0.5, 1, 5, 10, and 20 μM) did not affect cell growth on HUVECs.
ASM inhibitor 4i effectively reduces the content of Cers in liver and plasma in C57BL/6 male mice.
ASM inhibitor 4i shows antiatherosclerotic activity by inhibiting ASM, completely different from simvastatin.
ASM inhibitor 4i could lessen lipid plaques in the aortic arch and aorta and reduce plasma ceramide concentration and Ox-LDL levels.
ASM inhibitor 4i significantly decrease LPS-induced and Ox-LDL-induced cell inflammation by regulating the levels of ceramide and sphingomyelin.
