ATUX-1215 is a direct, orally active small molecule activator of protein phosphatase 2A (PP2A), suppresses bleomycin-induced fibrosis in vivo.
ATUX-1215 (1 uM) induces PP2A phosphatase activity responses in fibroblasts, reduced the level of TGFβ induction of ACTA2, COL1A1, COL1A2, and FN1 gene expression in fibroblasts.
ATUX-1215 activates PP2A responses in the BLM model and elicited potentially useful therapeutic effects.
ATUX-1215 (5 mg/kg, daily, oral) reduces the BLM-induced BALF levels of IL12p70, GM-CSF, and IL-1α in C57BL/6J mice.
ATUX-1215 shows the improved metabolic stability and bioavailability of ATUX-1215 to previously described PP2A activators.
ATUX-1215 prevents BLM-induced lung physiology changes, including the preservation of normal PV loop, compliance, tissue elastance, and forced vital capacity.
