Biological Activity
Auceliciclib (AU3-14, Ulecaciclib AU-314) is a potent and selective, brain-penetrant CDK4/6 inhibitor with Ki values ranging from 0.2 to 3.3 nM.
AU3-14 displaysselectivity that was 3-4 orders of magnitude greater for CDK4/cyclin D1 compared to CDK1/cyclin B1, CDK2/cyclin A2, CDK7/Mat1/cyclin H and CDK9/cyclin T1.
AU3-14 exhibits potent anti-proliferative effect against GBM cells with GI50 of2.17 - 3.54 µM.
AU3-14 inhibits Rb phosphorylation and arrests the cell cycle at the G1/G0 phase in GBM cells.
AU3-14 induces caspase-independent apoptosis in GBM cells. AU3-14 demonstrates potent tumor growth delay in GBM xenograft model.
