Biological Activity

AVI-4206 is a potent, specific small molecule inhibitor of SARS-CoV-2 ADP-ribosylhydrolase macrodomain (Mac1) with IC50 of 20 nM in HTRF assays.
AVI-4206 potently inhibits Mac1 with IC50 of 64 nM used auto ADP-ribosylated PARP10 and a coupled NudT5/AMP-Glo assay.
AVI-4206 engages Mac1 in cells with high specificity. an weakly inhibit viral replication in a gamma interferon- and Mac1 catalytic activity-dependent manner.
AVI-4206 reduces viral replication, potentiates innate immune responses, and leads to a survival benefit, in animal model of severe SARS-CoV-2 infection.
