Ac5GlutolNAc is a specific small molecule inhibitor of GlcNAc 2-epimerase (RENBP), selectively enhances the labeling efficiency of tetraacetyl-N-azidoacetylmannosamine (AAM) in antigen-presenting cells (APCs), but not non-APCs.
Ac5GlutolNAc (200-400 μM) significantly enhances AAM labeling efficiency without compromising the viability of DC2.4 cells.
Ac5GlutolNAc inhibits the flux from ManNAc toward GlcNAc and facilitates subsequent sialic acid biosynthesis.
Ac5GlutolNAc specifically amplifies AAM labeling efficiency in primary dendritic cells.
Ac5GlutolNAc interferes with the sialic acid synthetic pathways but not the GalNAc glycan synthesis pathways.
Ac5GlutolNAc amplifies AAM-mediated metabolic labeling of B cells and other immune cells in vivo.
Ac5GlutolNAc can enhance the labeling efficiency of AAM toward various types of APCs (dendritic cells, macrophages, and B cells).
