Amodiaquine (Amodiaquin, AQ) is an antimalarial agent and nuclear receptor Nurr1 (NR4A2) agonist, specifically binds to Nurr1-LBD (ligand binding domain) with EC50 of 20 uM.
Amodiaquine suppresses LPS-induced expression of proinflammatory cytokines (IL-1β, interleukin-6, TNF-α and iNOS) in a dose-dependent manner.
Amodiaquine significantly inhibits neurotoxin (6-OHDA-induced cell death in primary dopamine cells as examined by the number of TH+ neurons and dopamine uptake.
Amodiaquine exhibits neuroprotective effect in rat PC12 cells.
Amodiaquine also is a potent inhibitor of histamine N-methyltransferase protects mice from Propionibacterium acnes (P. acnes)-primed and lipopolysaccharide (LPS)-induced hepatitis.
