BKT300 is a small molecule inhibitor of leukemic cell migration and survival by targeting the PRC1 pathways, efficiently and specifically binds protein regulator of cytokinesis 1 (PRC1) with SPR Kd of 28.3 nM.
BKT300 does not bind to CDC25C or DCAF11.
BKT300 inhibits the migration of AML THP-1 cells toward monocyte chemotactic protein-1 (MCP-1).
BKT300 inhibits the formation and organization of actin filaments and microtubules.
BKT300 arrests cancer cells at G2/M and induces apoptotic cell death through the caspase 3 pathway, downregulates the expression of CDC25C but not CDC2 and upregulates p21.
BKT300 arrests PRC1 in a phosphorylated state at position T481.
BKT300 inhibits tumor growth and induces tumor regression in xenograft mouse models of AML.
