BPK-25 is a small molecule inhibitor of Nucleosome Remodeling and Deacetylation Complex (NuRD), suppresses T-cell activation at low-μM concentrations without causing cytotoxicity. BPK-25 inhibited TMEM173 activation by the cyclic dinucleotide ligand cGAMP. BPK-25 blocks NFAT activation, also reduces NFATc2 expression. BPK-25 promotes degradation of nucleosome remodeling and deacetylation (NuRD) complex proteins by a post-translational mechanism involving covalent protein engagement.
