BTdCPU is a potent activator of heme-regulated inhibitor (HRI) eIF2α kinase that promotes eIF2α phosphorylation and induces apoptosis in Dexamethasone-resistant cancer cells.
BTdCPU treatment induced phosphorylation of eIF2α and and upregulated mRNA and protein levels of the pro-apoptotic protein CHOP in Dex-resistant (MM1.R, RPMI 8266, U266) MM cells.
BTdCPU is cytotoxic for dex-resistant primary MM cells with relative sparing of normal cells.
BTdCPU triggers MCL-1 repression and synergizes with BH3-mimetics in mouse and human leukemia including both Ph+ and Ph-like B-ALL.
