BXY-14 is a highly potent TLR2 agonist, stimulates TNF-α release with EC50 of 2.2 nM and 1.8 nM in human THP-1 cells and mBMDCs respectively.
BXY-14 exhibited slightly stronger TLR2-activating activity than Diprovocim in hPBMCs, with EC50 values of 0.58 nM.
BXY-14 at 10 mg/kg induced stronger total IgG, IgG1, and IgG2c responses compared to Diprovocim in C57BL/6J mice.
At 10 mg/kg, BXY-14 in combination with the anti-PD-L1 antibody atezolizumab exhibited stronger tumor growth inhibition than Diprovocim.
BXY-14 monotherapy (20 mg/kg) potently suppressed tumor growth and prolonged survival, mechanistically linked to its downregulation of PD-L1 in tumors.
