Basmisanil (RG1662) is a potent, selective negative allosteric modulator of GABAA receptor α5-subtype with binding Ki of 5 nM for recombinant human GABAA-α5 receptors.
Basmisanil (RG1662) displays >90-fold selectivity versus α1, α2, and α3 subunit-containing receptors.
Basmisanil (RG1662) inhibits GABA-induced currents at GABAA-α5, yet has little or no effect at the other receptor subtypes.
Basmisanil (RG1662) attenuated diazepam-induced spatial learning impairment in rats (Morris water maze), improved executive function in non-human primates (object retrieval), without showing anxiogenic or proconvulsant effects in rats.
