CADD522 (CADD-522) is a small molecule that inhibits RUNX2-DNA binding (IC50=10 nM) and decreases RUNX2 transcriptional activity.
CADD522 had no effect on the RUNX2 occupancy in MCF7-Empty cells, CADD522 significantly inhibited RUNX2 enrichment in the MCF7-RUNX2 cells.
CADD522 reduced RUNX2-mediated increases in glucose uptake and decreased the level of CBF-β and RUNX2 phosphorylation at the S451 residue.
The administration of CADD522 into MMTV-PyMT mice resulted in significant delay in tumor incidence and reduction in tumor burden.
