CC0651 is a specific, allosteric inhibitor of ubiquitin-conjugating enzyme (E2) Cdc34 with IC50 of 1.72 uM for inhibition of ubiquitination of p27Kip1 by hCdc34. CC0651 has effect on activity of oher E2-E3 enzymes, including SMURF2, the Shigella IpaH enzyme SspH1, or the human RING domain enzyme Rnf168. CC0651 decreases cell proliferation and causes p27Kip1 accumulation in PC-3 cells. CC0651 engages a composite binding pocket formed from Cdc34A and ubiquitin. CC0651 also suppresses the spontaneous hydrolysis rate of the Cdc34A-ubiquitin thioester without decreasing the interaction between Cdc34A and the RING domain subunit of the E3 enzyme.
