Biological Activity

CD38 inhibitor 16 is a potent, selective and brain-penetrant CD38 inhibitor with IC50 of 2.71 nM and 1.94 nM for mCD38 and hCD37 respectively.
CD38 inhibitor 16 shows no inhibitory activity across a panel of off-target NAD+-consuming enzymes, consisting of sirtuins (SIRTs) and poly(ADP-ribose) polymerases (PARPs).
CD38 inhibitor 16 (3-100 mg/kg, oral) demonstrated dose-dependent increases in NAD+ levels in brain and lung tissues of mice.
