CMP05 (Endosomolytic compound CMP05) is a novel endosomolytic small-molecule compound that exhibits higher transfection efficiency/ability to induce endosomal escape of oligonucleotides (ONs) than current compounds such as chloroquine.
CMP05 had significantly higher efficacy than CMP01 at 5 uM, increased Splice-switching ON (SSO) activity.
CMP05 induced SSO leakage concurrently with endosomal swelling, induced SSO leakage in a mCherry-GAL9-recruitment-dependent manner.
CMP05 disrupted normal SSO trafficking through the endosomal network.
CMP05 was able to increase ON activity in a more efficacious manner than the previously published compound, UNC2383, in HeLa and HuH7 cell lines.
