Biological Activity
CN-0928 is a specific small molecule inhibitor of poly(C)-binding protein 2 (PCBP2), mitigates AD pathology by inhibiting PCBP2 condensation.
CN-0928 (1-2 uM) significantly reduced PCBP2 protein level, with the concomitant reduction of BACE1 but not APP or ADAM10 protein level.
CN-0928 (1 uM) significantly reduced PCBP2 condensates in SH-SY5Y-mCherry-PCBP2 cells.
CN-0928 significantly reduced protein levels of PCBP2 and BACE1 in the brain in animal model of 5×FAD mice, significantly reduced protein levels of CTFs as well as Aβ40/42 immunofluorescent signals.
CN-0928 binds to integrator complex subunit 1 (INTS1) and regulates PCBP2 transcription and protein level.
PCBP2 is an RNA-binding protein that contains KH domains and specifically binds the cysteine-rich motif, It is involved in cancer and iron metabolism.
