COH-SR4 (Mitochondria uncoupler SR4) is a bonafide uncoupler that induces energetic stress on cells, consequently leading to AMPK activation and mTOR inhibition without any effects on ERK/MEK MAPK signaling.
COH-SR4 prevents cell proliferation and promoted cell death in many human cancers including leukemia, lung cancer, melanoma, and hepatocarcinoma in vitro and in animal models.
COH-SR4 exhibits a strong inhibitory effect on the survival of melanoma cells (B16-F0 cell line IC50=5 uM, Hs600T cell lineI C50=6 uM, and A2058 cell line IC50=11 uM), does not cause any significant cytotoxicity in normal human aortic vascular smooth muscle cells (HAVSMC).
COH-SR4 inhibits vemurafenib-resistant melanoma in vivo.
