CPR1-B is a longer-acting phenylhexanamide mitofusin activator, increases mitochondrial motility in MFN2 T105M expressing mouse DRGs with EC50 of 3.3 nM (Emax=89%).
CPR1-B promotes the relaxed/active MFN2 conformation without affecting catalytic GTPase activity.
CPR1-B is a longer acting compound that can provide sustained mitofusin activation.
CPR1-B (60 mg/kg given orally twice daily) causes sustained mitofusin activation in 50-week-old MFN2 T105M mice.
CPR1-B modified phenotypes in vitro and in vivo in murine amyotrophic lateral sclerosis (ALS).
CPR1-B exhibits sustained brain levels, markedly prolonged pharmacodynamic effects, and disease modification in a proof-of-concept study of murine amyotrophic lateral sclerosis.
