CPUY192018 is a potent small-molecule inhibitor of the Keap1-Nrf2 PPI, has binding affinity for Keap1 with IC50 of 14.4 nM in FP assays and ITC Kd of 39.8 nM.
CPUY192018 activated the Nrf2-ARE pathway in vitro.
CPUY192018 stimulated the transcription of the Nrf2-ARE regulated cytoprotective genes in NCM460 cells in an Nrf2-dependent manner.
CPUY192018 produced cytoprotective effects against the DSS-induced injury in the NCM460 cells.
CPUY192018 (40 mg/kg/day, oral gavage) ameliorated the pathological symptoms in the DSS-induced mouse model of chronic ulcerative colitis.
