Biological Activity
CZ-2401 is a potent, specific inhibitor of the TRIM47-HNF4α interaction, binds to HNF4α with MST Kd of 2.52 uM, stabilizes HNF4α protein and activates HNF4α transcriptional activity.
CZ-2401 enhances HNF4α stability by blocking HNF4α–TRIM47 interaction.
CZ-2401 exhibits potent cytotoxic effects in both Huh7 and Hep3B cells but a minimal cytotoxicity in HEK293T cells with absent HNF4α expression.
HNF4α knockdown in Huh7 and Hep3B cells substantially attenuated CZ-2401-induced cytotoxicity, confirming that CZ-2401 specifically targets HNF4α.
CZ-2401 (8 mg/kg, i.p.; Vehicle: saline solution) ameliorates the TRIM47-driven HCC progression in mouse xenograft model by subcutaneously inoculating nude mice with Hep3B cells.
