Cevidoplenib dimesylate (SKI-O-703) is a potent, selective, orally active spleen tyrosine kinase (Syk) inhibitor with IC50 of 6.2 nM. SKI-O-703 is the orally bioavailable dimesylate salt form of Cevidoplenib (SKI-O-592, Cat#PC-61366). SKI-O-592 is approximately 9-fold more potent than R406 as a Syk kinase inhibitor. SKI-O-592 is highly specific for Syk, displays 67- to 2753-fold selectivity against a panel of kinases. SKI-O-703 significantly ameliorated synovitis, with fewer neutrophils and macrophages infiltrated into the synovial tissue, in model of serum-transferred arthritis. SKI-O-703 attenuates the progression of autoantibody-mediated autoimmune diseases by inhibiting both autoantibody-producing and autoantibody-sensing cells.
