Biological Activity
Compound 150441 is a highly specific small molecule inhibitor of Cdc2-like kinase 4 (CLK4) with IC50 of 21.4 nM.
Compound 150441exhibits weaker inhibitory activity against CLK1, CLK2, and CLK3, as well as against a panel of 60 kinases.
Compound 150441 (10 uM) exhibits significant cytotoxicity (IC50<1 uM) and more than 95% anti-proliferative effects in both Mia PaCa-2 and Panc-1 cell lines.
Compound 150441 affects SR protein activity and subsequently influence the RNA splicing process.
Compound 150441 at 3 and 10 uM led to a substantial downregulation of the anti-apoptotic proteins Mcl-1 and Bcl-2, along with the activation of the caspase-dependent apoptotic pathway in Mia PaCa-2 and Panc-1 cell lines.
