D-e-MAPP (D-erythro-MAPP) is a ceramide analogue and specific inhibitor of alkaline ceramidase, inhibits alkaline ceramidase activity in vitro with IC50 of 1-5 uM.
D-e-MAPP does not modulate the activity of other ceramide metabolizing enzymes in vitro or in cells, and is a poor inhibitor of acid ceramidase (IC50>500 microM).
D-e-MAPP treatment elevates endogenous levels of ceramide with the consequent biologic effects of growth suppression and cell cycle arrest in HL-60 human promyelocytic leukemia cells.
D-e-MAPP induces apoptosis and suppression of proliferation in HaCaT keratinocytes.
