Biological Activity
D347-2761 is a small molecule c-Myc inhibitor, blocks c-Myc/Max heterodimer and disturbs c-Myc protein stability simultaneously, inhibits c-Myc-mediated transcriptional activities.
D347-2761 dose-and time-dependently inhibited RPMI-8226 and NCI-H929 cell viabilities significantly, but not normal PBMCs.
D347-2761 synergized with bortezomib to promote DNA damage in RPMI-8226 and NCI-H929 cells.
D347-2761 overcame BTZ drug resistance in MM cells.
D347-2761 inhibited primary tumor growth in humanized myeloma xenograft mouse model.
D347-2761 disturbed c-Myc stability to regulate downstream target genes, inhibited the expression of ACAT1, CDK4, NME1, PIM3, and SMYD2 at the mRNA levels in RPMI-8226 cells.
