DDO-6079 is a specific, allosteric small-molecule CDC37 inhibitor, binds to an allosteric site on the CDC37 M-terminal domain with ITC Kd of 1.03 uM, inhibits the binding between HSP90-CDC37 and CDC37-kinase.
DDO-6079 simultaneously inhibits HSP90-CDC37 and CDC37-CDK4/6 chaperone comple.
DDO-6079 inhibits the formation of the HSP90-CDC37-CDK4/6 complex in cells in an allosteric manner and selectively inhibits the maturation of multiple oncogenic kinases escape from the heat shock response (HSR).
DDO-6079 exhibits no effects on HSR related protein (HSF-1, HSP70, HSP40 or HSP27).
DDO-6079 shows antiproliferation activity and decreased the thermostability of CDK6 to reverse the resistance of colorectal cancer cells to palbociclib.
DDO-6079 reversed the resistance of CDK6 to palbociclib (a successful CDK4/6 inhibitor) in colorectal cancer cells and exhibited efficacy in vivo.
