DEPTOR-IN-1 (Compound C3) is a small molecule putative DEPTOR inhibitor with MST KD of 9.3 uM, exhibits antimultiple myeloma activity with IC50 of 1.09 uM for RPMI 8226 cells. DEPTOR-IN-1 induces RPMI 8226 apoptosis, and blocks cell cycle in the S phase. DEPTOR-IN-1 displays satisfactory inhibition of tumor growth in RPMI 8226 xenografts without obvious side effects. DEPTOR is a 48kDa protein that binds to mTOR and inhibits this kinase within mTORC1 and mTORC2 complexes. DEPTOR inhibits mTORC1 and mTORC2 to induce autophagy.
