Biological Activity

DJ503701 (compound 11) is a small molecule ligand competition binding at FZD6, efficiently counteractes WNT-3A-induced WNT–β-catenin signaling with IC50 of 1.2 uM.
Compound 11 binds nonselectively to the receptor core of FZDs.
Compound 11 targets the transmembrane core of FZDs and not the orthosteric WNT binding site on the cysteine-rich domain (CRD).
Compound 11 interferes with WNT-driven signaling in hepatocyte spheroids and pancreatic cancer cells.
Compound 11 reduces viability of RNF43-negative pancreatic cancer cells.
