Biological Activity
DLG-41 is a potent, selective inhibitor of GAS41 YEATS domain with IC50 of 610 nM in in AlphaLISA assays, binds to GAS41 YEATS domain with ITC KD of 1.0 uM.
DLG-41 blocks binding of GAS41 with chromatin in HEK293T cells with IC50 of 0.9 uM, with no activity for BRD4-BD1.
DLG-41 blocks proliferation of a panel of NSCLC cell lines in a GAS41 dependent manner (H1299 cell IC50=0.45 uM).
DLG-41 decreases global gene expression and ameliorates NSCLC phenotypes in H1299 cells, associated with lung cancer cell identity, tumor migration, and invasion.
