Dual GO/LDHA inhibitor 2 is a first-in-class, orally bioavailable dual inhibitor of glycolate oxidase (GO) and lactate dehydrogenase A (LDHA) with Ki of 390 nM and 40 nM, also promotes hydrophobic tag-mediated autophagic degradation of LDHA.
Dual GO/LDHA inhibitor 2 (20 mg/kg/day) degrades hepatic LDHA, decreases urinary oxalate, and diminishes renal deposition of calcium-oxalate crystals in the liver of PH1 mice.
Dual GO/LDHA inhibitor 2 (20 mg/kg body weight) exhibits a significant reduction in urinary oxalate levels and urinary glycolate levels at the renal level of PH1 mice.
